Antiphospholipid antibody (25µg/mL) treatment for 16h. Untreated controls were given media only.

Placental explants were cultured in Advanced DMEM/F12 media containing 2% FBS and 1%PS for 16h, at 37°C and 5%CO2.

RNA was extracted using Trizol and the Invitrogen PureLink mini kit according to manufacturer's instructions.

Biotinylated cRNA was prepared according to the standard Affymetrix protocol from 1µg total RNA (One-cycle cDNA synthesis).

Following fragmentation, 5ug of cRNA was hybridized onto Affymetrix Human Genome U133 Plus 2 GeneChips. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the Affymetrix GeneChip Scanner 3000 7G system.
