Extraction protocol |
Phenol-chloroform extraction was used to isolate total RNA, which was enriched using the Ambion MicrobExpress kit. All RNA samples were tested for integrity on a BioRad Experion capillary electrophoresis system. Possible residual DNA was removed by addition of Ambion Turbo DNase. Library preparation for Applied Biosystems SOLiD sequencing was done according to the manufacturer’s Whole Transcriptome Library Preparation protocol.

Raw color space data from SOLiD sequencing was mapped to the Bacillus anthracis Ames Ancestor genome (NC_007530.2, NC_007322.2, NC_007323.3) using SOCs software with a mismatch cutoff of 5 nucleotides.
