Treatment protocol |
8093 cells were treated for 20 days with 20 nM nilotinib, or for 26 days with 1 uM lonafarnib. Bin-2 cells were treated for 21 days with 50 nM nilotinib, or for 30 days with 0.25 uM lonafarnib. These drug concentrations were earlier determined in pilot experiments to cause most of the leukemia cells to die, but allow a small percentage to grow out at the end of the treatment period as cells that had become drug-tolerant to the concentration of that drug used. Fresh drug was added with changes of fresh medium, generally every second day. For isolation of RNA at the t= 0 and t= end/resistance points we used a 10-cm dish per biological replicate. For the t= midpoint one 15-cm dish or two pooled 10-cm dishes were used to obtain sufficient cells.

Mouse preB ALL cells were grown on irradiated mouse embryonic fibroblast feeder layers in McCoy's 5A medium supplemented with 15% FCS, 110 mg/L sodium pyruvate, L-glutamine, penn/strep, 10 ng/ml recombinant IL-3 and 50 uM beta-mercaptoethanol.
