We euthanized mice by asphyxiation with CO2, harvested their testes, and isolated Sertoli cells as previously described [Chang et al., 2011; Anway et al., 2003]. We extracted RNA from purified Sertoli cells (pooled from five to seven mice) by using a Qiazol miRNeasy kit (Qiagen, Valencia, CA). We converted 1 μg of total RNA into cDNA by using a MiScript cDNA synthesis kit (Qiagen) and analyzed with quantitative real-time RT-PCR using miRNA-specific primers. We normalized miRNA expression levels to RNU19/5S and gene expression to 18S.
