Total RNA was prepared in accordance with Illumina's sample preparation protocol for PE sequencing of mRNA. mRNA from total RNA was isolated using poly-T oligo-attached magnetic beads and fragmented with fragmentation buffer in mRNA Sequencing Sample Prep Kit (Illumina). Following steps were identical with exome sequencing, except that purified fragments size from agarose gel was 300bp, bigger than exome sequencing.
