SRX7389890: Bacillus Transcriptome analysis

Design: After removing the rRNA, the total RNA was interrupted to short fragments and the first-strand cDNA was synthesized with random hexamer-primer. Then the second-strand cDNA was synthesized with dATPs,dGTPs,dCTPs,dUTPs, RNase H and DNA polymerase I. The obtained fragments were purified with QiaQuick PCR extraction kit, repaired the end, added poly(A), connected the sequencing adapters and degraded the second-strand cDNA with UNG enzyme. Finally, the resulting fragment was purified by MiniElute PCR Purification Kit before PCR amplification.
