Manufacture protocol
1. Use the primer 5.0 to design probes and test them with oligo6.0 software. The principles of probe design as follows:(1) the length of probe is 59 bases, and annealing temperature is 78.0±5.°C, (2) tandem duplication bases was not more than 6,(3) the stem structure of stem loop in the internal of probes was not more than 6bases, (4) the most stable secondary structure base pairing of probe molecule was less than 6bp, (5) the Score value is less than 40, after comparing the probes with non-targeting gene sequence, which means the serial homology fragments were not more than 20 bases. 2. Dissolve the probes to 40 uM solution with sterilization, and inject into 169 pore plate after mixed with sample application buffer solution. Then put the probes on the surface of amination film base (home made) by using GeeDomSmartArrayer 48.
