A Yeast Genome Oligo Set consisting of 5’ amino acid-modified 70-mer probes was purchased from Operon. Then Oligonucleotides were dissolved in EasyArrayTM spotting solution (CapitalBio Corp) at 40 uM concentration, and printed on PolymerSlide (75×25 mm), in which the surface was covered by a thin layer of aldehyde group modified three-dimensional polymer chain (CapitalBio Corp.). A set of microarray containing a total of 23184spots, including 6400 clones in the form of PCR products and controls: Hex was used as mobilized positive control; DMSO was used as spot solution and negative control; Five housekeeping genes of yeast genome, ACT1, RPL32, SSA4, TUB4, UB14 were used as positive controls, and Eight sequences derived from intergenic regions of the yeast genome were used as external controls. These sequences were PCR amplified and cloned into plasmid pSP64Poly (A) (Promega) to produce poly (A)-RNA after in vitro transcription. All of the PCR products and controls were spotted in triplicate on slide. On the slide, there are 48 blocks and every block has 21 columns and 22 rows.
