Summary |
We obtained Nab3, Nrd1, and RNA polymerase II occupancy profiles across the genome of S.cerevisiae in wild-type and rrp6∆ strains. This allowed us to determine the impact of defective nuclear exosome on NNS-dependent transcription termination.

Overall design |
RNA was crosslinked to Nab3, Nrd1, or RNA polymerase II and sequenced after protein purification under stringent and denaturing conditions. This reveals the position of the proteins at the nucleotide resolution level and the extent of their binding to RNA in wild-type versus rrp6∆ cells.
