Summary |
We mapped the genome-wide binding profiles of GR by using ChIP-Seq in livers from mice fed control or HFD diet after acute exogenous ligand (DEX) administration.

Overall design |
Male C57BL/6J mice were fed either control diet (11 kcal% fat w/sucrose, Research Diets D12329) or High Fat Diet (58 kcal% fat w/sucrose, Research Diets D12331) for 12 weeks. At 17 weeks of age, Dexamethasone (Sigma, D2915) was administered at ZT0 and at ZT12 as a single i.p. injection of 10mg/kg. Mice were sacrificed by cervical dislocation 1h later. ChIP was performed as previously described (Uhlenhaut et al. 2012). Sheared chromatin was immunoprecipitated with two different anti-GR antibody (Santa Cruz, sc-1004X and ProteinTech, 24050-AP). Two replicates per condition and time point were used, for a total of 12 GR (ZT0,ZT12, control/HFD) ChIP-seq samples.
