Treatment protocol |
Male C57BL6/J Rj mice were obtained from Janvier (France) and were randomly assigned to rapamycin or vehicle treatment groups. We had three treatment cohorts, distinguished by age of treatment onset. Treatment was initiated in young adulthood (i.e., at approx. 4 months of age), in midlife (i.e., at 13 months of age) or late in life (i.e., at 20-22 months of age). Animals were treated for >12 months before analyses started at 16 months, 25 months and 34 months of age, respectively. Animals were kept on their respective diet for the entirety of the study. In all cases, the experimenter was blinded to the animal group assignments.

Extraction protocol |
RNA was extracted with Trizol reagent, followed by clean-up with QIAGEN RNeasy midi kit in accordance with the prescribed protocol provided with the kit. Quality control was performed with gel electrophoreses.
