Treatment protocol Atopic subjects underwent bronchoscopies before and 48 h after an allergen challenge. The same procedure was repeated on each donor, 1 month after an injection with mepolizumab (anti-IL-5) that decreases the amount of airway eosinophils after allergen challenge.
Growth protocol No growth, BAL cells are used immediately after broncoscopy

Extraction protocol Trizol extraction of total RNA from BAL cells was performed according to the manufacturer's instructions
Label biotin
Label protocol cDNA were prepared using Ambion whole transcript target labelling kit according to the manufacturer's instructions. Fragmentation and labeling of the single-stranded cDNA was performed using the Affymetrix GeneChip WT Terminal Labeling Kit (Affymetrix, Santa Clara, CA, USA).
 
Hybridization protocol The labeled cDNAs were hybridized to Human Gene ST 1.0 GeneChips (Affymetrix) according to the manufacturer’s protocols.
Scan protocol GeneChips were scanned using the GeneChip® Scanner 3000 (Affymetrix).
Description Gene expression in BAL before allergen challenge and 1 month after mepolizumab injection
Data processing Raw data (CEL files) were uploaded into ArrayStar software version 4.0 (DNASTAR Inc., Madison, WI, USA) for normalization and statistical analysis. The robust multichip analysis (RMA) algorithm was used for background correction, quantile normalization and median polish summarization.
