Total RNA was isolated from equal amounts of secretion samples collected from BPH and PCa patients using ”mirVana PARIS kit” (Ambion, Darmstadt, Germany) according to the manufacturer’s protocol.

50 ng of total RNA from each sample are labeled with Cy3, via using “Agilent miRNA Labeling and Hyb Kit”.

Hybridization protocol |
Labeled RNAs are heat denatured and hybridized to Agilent 8x60K miRNA microarray V4 comprised of 1349 probes targeting a comprehensive selection of 1205 human and 144 virus miRNAs with control probes from Sanger miRBase (release 16) at 55o C for 20 hr.

Scan protocol |
After hybridization and post-hybridization washes, the chip was scanned by Agilent Microarray Scanner with Surescan High Resolution Technology (Agilent Technologies, Santa Clara, CA).

Data processing |
Scanned images were analyzed with Feature Extraction v10.7.3.1 (Agilent Technologies, CA) and Bioconductor software. Data are normalized by quantile normalization. P values and fold changes are calculated.
