Cells were infected with pLenti6.3-RNF2WT or pLenti6.3-GFP lentiviruses, selected with blasticidine (5mg/ml) till control uninfected cells were dead.

Cells were maintained in standard tissue culture conditions (5% CO2 , 37oC).

RNA was isolated using the RNeasy Kit per manufacturer's instructions. cDNA was prepared using SuperScript III using the manufacturer's instructions. qPCR were performed using SybrGreener (Invitrogen) and Stratagene instrument.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on Affymetrix Human Genome U133 Plus 2.0 Arrays. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

Scan protocol |
GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G2500A.

Microarray data was normalized by using RMA normalization with the Bioconductor affy package. Data was analyzed using the LIMMA Bioconductor package.
