Extraction protocol |
Total RNA was extracted using the RNeasy Mini Kit(Qiagen, MD, USA). The concentration was measured by ND-1000 UV-VIS spectrophotometer(NanoDrop Technologies, DE, USA). The quality of purified RNA(RNA integrity number, RIN) was detected by the RNA 6000 Labchip Kit and Agilent 2100 Bioanalyzer(Agilent, CA, USA).

Initially 100ng of total RNA was added into the reaction to generate 1.65µg Cy3-labeled cRNA using Low Input Quick Amp Labeling Kit(Agilent).

Hybridization protocol |
cRNA was hybridized to Agilent 4×44K Whole Human Genome microarray.

Scan protocol |
Slides were washed and then scanned with the Agilent G2505B Microarray Scanner System. Fluorescence intensities of the scanned images were extracted and preprocessed with the Agilent Feature Extraction Software (v9.1).
