Treatment protocol |
To load the Signal 1 (S1), THP-1 cells were incubated for 3 hours in the culture medium with or without inclusion of 0.2 microgram/ml lipopolysaccharide (LPS). To load the Signal 2 (S2), they were incubated further for 2 hours in the culture medium supplemented with 10 microM nigericin sodium salt dissolved in ethanol or the equal v/v% concentration of ethanol (vehicle).

Growth protocol |
THP-1 cells provided by Riken Cell Bank were maintained in RPMI 1640 medium supplemented with 10% fetal bovine serum (FBS), 55 microM 2-mercaptoethanol, 2 mM L-glutamine, 100 U/ml penicillin and 100 microgram/ml streptomycin (culture medium).

Extraction protocol |
Total cellular RNA was isolated by using the TRIZOL Plus RNA Purification kit (Invitrogen). The quality of total RNA was evaluated on Agilent 2100 Bioanalyzer (Agilent Technologies).

Three hundred ng of total RNA was processed for cDNA synthesis, fragmentation, and terminal labeling with the GeneChip Whole Transcript Sense Target Labeling and Control Reagents (Affymetrix).

Hybridization protocol |
Hybridization was performed at 45℃ for 17 hours. The arrays were washed and stained in the GeneChip Fluidic Station 450 (Affymetrix) by using Hybridization, Wash and Stain kit (Affymetrix).

Scan protocol |
The arrays were scanned by the GeneChip Scanner 3000 7G (Affymetrix).

The data expressed as CEL files were normalized by the robust multiarray average (RMA) method with the Expression Console software (Affymetrix).
