Extraction protocol Mice between 14 and 16 weeks old were fed 2 g/kg doxycycline to induce Celf2 expression in heart. All food was removed from cages 2-3 hours prior to addition of dox food. At the indicated time point, the mouse was sacrificed, and the heart was dissected. Blood was removed by washing with PBS. The atria were removed, and small portion of the apex was removed. The remaining ventricle tissue was frozen in liquid nitrogen. Total RNA from the ventricle tissue was prepared by lysis in guanidium isothiocyanate, followed by phenol/chloroform extraction, and prepared for RNA sequencing.
Poly(A)+ RNA was selected by oligo-dT magnetic beads, and prepared for paired end RNA sequencing using Illumina paired end sequencing primers.

Data processing Reads were aligned to the mm9 mouse genome and splice junctions using tophat/bowtie.
