Sample GSM1593861 Query DataSets for GSM1593861
Status Public on Jan 29, 2015
Title Nax knockdown HaCaT high sodium concentration 16 hour stimulation_2
Sample type RNA

Source name keratinocyte cell line
Organism Homo sapiens
Characteristics gender: female
tissue: skin epithelium
cell line: HaCaT
genotype/variation: Nax knockdown
treatment: high sodium concentration
time: 16 hour
Treatment protocol A treatment with control medium (final150 mM sodium) or medium plus 10% more sodium (final 165 mM sodium) was applied to each well. Cells were harvested at time point 0, 4, and 16 hours post treatment.
Growth protocol Both wild type and Nax knockdown HaCaT cells were seeded in 12-well plates. 70-80% confluent cells were starved with fetal bovine serum (FBS) free Dulbecco's modiļ¬ed Eagle's medium (DMEM) overnight
Extracted molecule total RNA
Extraction protocol The total RNA from wild type and Nax knockdown HaCaT cells
Label biotin
Label protocol Single-stranded cDNA was generated from the amplified cRNA with the WT cDNA Synthesis Kit (Affymetrix) and then fragmented and labeled with the WT Terminal Labeling Kit (Affymetrix).

Hybridization protocol Hybridization performed at the Interdisciplinary Center for Biotechnology Research Microarray Core, University of Florida. The microarray chips were Affymetrix GeneChipĀ® Human Transcriptome Array 2.0
Scan protocol Array scanning was performed according to the manufacturer's instruction (Affymetrix)
Description Sample name: HT_Nx_16h_2
Data processing The microarray data was processed with Affymetrix Expression Console and Transcriptome Analysis Console (Affymetrix).
RMA expression value derived from Expression Console software; core-exon analysis
