Treatment protocol Bioreactor culture was performed in the presence of mesh A (Parietex) or mesh B (ULTRAPRO).
Growth protocol Fibroblasts were generated by collagenase digestion for 16h in DMEM+10% fetal bovine serum (FBS)+1% Penicillin/Streptomycin (Pen/Strep) at 37°C, after sieving, life cells were precultured in DMEM+10% FBS+1%Pen/Strep; for bioreactor culture, 750,000 cells were embedded in 5ml collagen type 1 gel matrix, fed with 5ml DMEM+10% FBS+1%Pen/Strep and incubated for 14 days at 37°C.
Extracted molecule total RNA
Extraction protocol Total RNA was isolated using the miRNeasy kit (Qiagen) according to the manufactuer's instractions.
Label biotin
Label protocol 100ng of total RNA were amplified and labeled with the IVT Express Kit (Affymetrix); all steps were performed according to the manufacturer's protocols.
 
Hybridization protocol 130µl hybridization mix prepared from labeled cRNA fragments was hybridized for 16h at 45°C and 60 rpm to GeneChip PrimeView arrays. Microarrays were washed and stained in the Affymetrix Fluidics Station 450 using script FS450_0002.
Scan protocol Microarrays were scanned using a GeneChip Scanner 3000 7G (Affymetrix).

Data processing Data were preprocessed in the R v3.0.1 environment with Bioconductor package affy; background correction, normalization and probeset summary were performed with the rma command using default parameter settings.
