Status Public on Jul 23, 2015
Title C4 13C
Sample type RNA

Source name stable isotope incubations of Monterey Bay surface water
Organism marine metagenome
Characteristics tissue: surface marine microbes (<1 micron) collected from Monterey Bay seawater
incubation condition: glucose, 36 hours
Treatment protocol samples included \ 13C labeled glucose, starch, acetate, lipids, protein, or ammino acids added at 1 micromolar C final concentration
Growth protocol 1 micron filtered samples incubated in 12 Liter bottles at 20 degrees Celsius for 12 hours or 36 hours
Extracted molecule total RNA
Extraction protocol RNA was extracted using the Qiagen Rneasy kit
Label 13C
Label protocol samples 1 and 2 were labeled with Alexafluor 532 with the Ulysis labeling kit from Life Technologies

Hybridization protocol 1 microgram RNA was hybridized with Nimblegen Hybridization buffer at 42 degrees Celsius for 18 hours, and washed with Nimblegen Wash buffers I, II, and III
Scan protocol samples 1 and 2 were scanned with a Genepix 4000B at PMT = 800, other samples scanned with a NanoSIMS 50 according to protocol from Mayali et al. (2012) ISME J 6:1210-1221.
Data processing Isotope ratios extracted using software package LIMAGE (L. Nittle, Carnegie Institute, Washington DC)
