Growth protocol |
Lineage negative bone marrow cells were isolated from C57B6/J mice. Cells were cultured in StemSpan SFEM with 15% FBS, 10 ng/ml rmIL-3, 50 ng/ml rmSCF, 50 ng/ml rmFLT3L, 1% penicillin-streptomycin, and 1% glutamine, at the concentration of 2 x 10^5 cells/ml, and prestimulated overnight. Cells were retrovirally transduced with control MigR1 (Mig) or MigR1-HA-RUX1-ETO (RE) for 24 hours. After transduction, cells were washed, resuspended in StemSpan SFEM with or without addition of 10ng/ml of rmGM-CSF for 24 hours. After a total of 48 hours of RUNX1-ETO expression and 24 hours of GM-CSF treatment, GFP+/Lineage-/c-Kit+ cells were sorted for RNA isolation.

RNA was isolated using column purification with the QIAGEN RNeasy mini kit in accordance with the protocol provided with the kit. Quality control was performed with Agilent Bioanalyser.
