Treatment protocol |
Cells were sorted from mouse whole bone marrow, treated with DMSO for 2 hours, then transplanted into primary recipients. Cells were isolated from primary recipients one year later then transplanted into secondary recipients, without further treatment. After 3 months, cells were isolated from secondary recipients and used for total RNA extraction.

Amplification and library preparation was performed using the Ovation Pico WTA System V2 and Encore Biotin Module labeling.

Samples were hybridized to Mouse Gene 2.0 ST Arrays (Affymetrix) and scanned at the Microarray Core Facility at the IDDRC of Boston Children's Hospital and Harvard Medical School.

Data processing |
Raw data were processed using the Affymetrix Command Console Software to determine RMA-normalized expression values (log2) and used to identify differences in gene expression between GFP (+) and (-) samples.
