Primary mouse articular chondrocytes were infected with null(control) or recombinant adenovirus expressing Hypoxia inducible factor-2 alpha (HIF-2α). The cells were harvested for total RNA extraction at 24 h post infection.

Cultured were maintained at 37 °C in an humidified atmosphere containing 5% CO2 as a monolayer in Dulbecco’s modified Eagle’s medium (DMEM; Gibco) supplemented with 10% fetal bovine serum and antibiotics (penicillin G and streptomycin). 37 °C, 5% CO2

Extraction protocol |
Total RNA was extracted from mouse articular chondrocytes using a Purelink RNA mini kit (Ambion). The concentration, purity, and integrity of the extracted RNA were determined by spectrophotometry.
