Treatment protocol Primary mouse articular chondrocytes were infected with null(control) or recombinant adenovirus expressing Hypoxia inducible factor-2 alpha (HIF-2α). The cells were harvested for total RNA extraction at 24 h post infection.
Growth protocol Cultured were maintained at 37 °C in an humidified atmosphere containing 5% CO2 as a monolayer in Dulbecco’s modified Eagle’s medium (DMEM; Gibco) supplemented with 10% fetal bovine serum and antibiotics (penicillin G and streptomycin). 37 °C, 5% CO2
Extracted molecule total RNA
Extraction protocol Total RNA was extracted from mouse articular chondrocytes using a Purelink RNA mini kit (Ambion). The concentration, purity, and integrity of the extracted RNA were determined by spectrophotometry.
Label biotin
Label protocol The sense cDNA was then fragmented and biotin-labeled with TdT (terminal deoxynucleotidyl transferase) using the GeneChip WT Terminal labeling kit
 
Hybridization protocol Approximately 5.5 μg of labeled DNA target was hybridized to the Affymetrix GeneChip Array at 45°C for 16hour
Scan protocol Hybridized arrays were washed and stained on a GeneChip Fluidics Station 450 and scanned on a GCS3000 Scanner (Affymetrix).

Data processing Array data export processing and analysis was performed using Affymetrix® GeneChip Command Console® Software (AGCC). Probe signals in the raw data were normalized by the RMA (Robust Multi-array Average)
