Male mice with a floxed Snord116 deletion (B6(Cg)-Snord116tm1Uta/J - JAX stock no 008118) were stereotaxically injected with AAV-Cre or AAV-GFP in the mediobasal hypothalamus and brains were fresh frozen at 10 weeks post injection.

Hypothalamic regions were laser-capture microdissected from cryosections using a P.A.L.M Microlaser system, and RNA extracting using the miRNeasy micro RNA extraction kit (Qiagen). Amplification and cDNA library prepartion was performed using the Pico Input Mammalian SMARTer Stranded RNA-Seq kit (Takara Bio).
