Treatment protocol |
Lambs were euthanized, semimembranosus muscle excised and fixed in RNAlater (Ambion).

Lambs were housed indoors and allowed to nurse ewes fed grain based diet until collection.

Extraction protocol |
Tissue homogenized in 4M guanidinium thiocyanate, 25mM EDTA, 1% sodium-N-lauroyl-sarcosine. Homogenates ultracentrifuged on 5.7M CsCl cusion. RNA purified and DNase treated with Nucleospin RNA II columns (Machery-Nagel Inc.).

Biotinylated cRNA was prepared according to the standard Affymetrix protocol from 5 micrograms total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 20 micrograms of cRNA were hybridized for 16hr at 45C on GeneChip Bovine Expression Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

Scan protocol |
GeneChips were scanned using the GeneChip Scanner 3000 7G.

Data processing |
Images were interpreted with Microarray Suite version 5.0 (MAS5.0) in GCOS with no scaling or normalization. Images were also interpreted by RMA using Bioconductor package of R. The VALUEs below represent RMA signals.
