Treatment protocol |
Mononuclear cells were separated by gradient centrifugation using Ficoll-Hypaque from cord blood and postnatal peripheral blood samples within 24 h of the sample collection.
Extracted molecule |
total RNA |
Extraction protocol |
DNA extracted by Qiagen AllPrep kit.
Label |
Cy3
Label protocol |
Starting with 100 ng of the isolated RNA for each sample, double stranded cDNA and cyanine 3 labeled cRNA were synthesized using low input quick amp labeling kit, one-color and RNA spike-in kit, one-color (Agilent).
Hybridization protocol |
The resultant cRNAs were fragmented for 30 min at 60°C in dark. The fragmented cRNAs (600 ng) were hybridized on the microarray at 65°C for 17 h.
Scan protocol |
After washing, slides were scanned with the Agilent microarray scanner G2505B.
Data processing |
Feature extraction software (version 10.7.3.1) was used to calculate signal intensities from the scanned images.
