Extraction protocol Phenol chloroform extraction of human brain tissue. Genpathway Inc performed the following: DNA (12 ug) was precleared with protein G agarose beads (Invitrogen). Methylated DNA was detected using a sheep polyclonal antibody against 5-methyl-cytosine. After incubation at 4C overnight, protein G agarose beads were used to isolate the immune complexes. Complexes were washed and eluted from the beads with SDS buffer. IP’ed DNA was purified by phenol/chloroform extraction and ethanol precipitation.
