Extraction protocol |
Argonaute complexes were immunoprecipitated with beads coated with anti-Ago1 or anti-Ago2 complexes. RNA was isolated by adding 40 μg Proteinase K in 200 μl Proteinase K buffer (300 mM NaCl, 25 mM EDTA, 2% SDS, 200 mM Tris HCl pH 7.5) directly onto beads followed by Phenol/Chloroform extraction and Ethanol precipitation. Small RNA cloning was performed by Vertis Biotechnologie AG (Weihenstephan, Germany)
