Treatment protocol The gene was overexpressed in SAS-3.4 cells and cultured for 48 hours. After 48 hours, total RNA of SAS cells was extracted.
Growth protocol SAS cells were obtained from the Japanese Collection of Research Bioresources Cell Bank. SAS cells were cultured in Dulbecco’s modified Eagle medium (DMEM; Invitrogen, CA, USA) supplemented with 10% heat-inactivated FBS, 1% AA and 10mg/mL Puromicine at 37 °C in 5% CO2.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted using QIAzol and the miRNeasy Mini Kit (Qiagen, Hilden, Germany) according to manufacturer’s protocols. RNA was quantified using a NanoDrop-1000 spectrophotometer and a Agilent 2100 bioanalyzer (Agilent Technologies, Palo Alto, Calif.).
