Extraction protocol |
Cryopulverization of Kidney tissue using the Biopulverizer and homogenization using the Mini-Bead-Beater-16 . RNA was prepared using the TRIzol® Reagent (Invitrogen life technologies) following the manufacturer's recommendations. RNA was quantified and quality monitored using a NanoDrop-1000 spectrophotometer and the Baseline-ZERO DNase(EPICENTRE, Cat. Nos. DB0711K).

Cyanine-3 (Cy3) labeled cRNA was prepared from 1 ug RNA using the Quick Amp Labeling Kit, One-Color (Agilent p/n 5190-0442) according to the manufacturer's instructions, followed by Purify the labeled/amplified RNA and labeled cRNA QC. Take 1.5 μl of each sample to determine the yield and specific activity by using the NanoDrop ND-1000.

Hybridization protocol |
1.65 ug of Cy3-labelled cRNA (the yield is >1.65 μg and the specific activity is >9.0 pmol Cy3 per μg cRNA) was fragmented at 60°C for 30 minutes following the manufacturers instructions using Agilent Gene Expression Hybridization Kit (Agilent p/n 5188-5242). On completion of the fragmentation reaction, hybridize at 65°C for 17 hours After hybridization, microarrays were washed.

Scan protocol |
Slides were scanned immediately after washing on the Agilent Microarray Scanner (Agilent p/n G2565BA) using one color scan setting (Scan Area 61x21.6 mm, Scan resolution 5um, Dye channel is set to Green and Green PMT is set to 100%).
