Growth protocol |
For differentiation, iPSCs were plated on Matrigel at 3.15e5 cells/cm2. Differentiations were initiated 24 hours after plating and were carried out in X-VIVO10 (Lonza 04-743Q). Factors were added at the following concentrations: Activin A, 100ng/ml (d0-4); Wnt3a, 50ng/ml (d0, and d9-13); TTNPB (RAR agonist) 6nM (d4-13); BMP4, 20ng/ml (d5-13); LY364947, 5M (d5-13); FGF8b or FGF8a, 50ng/ml (d9-13); SAG, 100ng/ml (d5-8); SANT-1, 0.25M (d9-13) insulin-transferrin-selenium, 1:5,000 (d0) or 1;2,000 (d1-4). At day 20, cultures where transplated into athymic nude mice under the kidney capsule. Grafts were harvested 14-17 post transplantation and prepared for either bulk or scRNA-seq.

RNA was extracted using Qiagen RNeasy Minikit (Cat No./ID: 74106). RNA was then submitted to the Genomics and Microarray Shared Resource Core at the University of Colorado Anschutz Medical Campus RNA libraries were prepared for sequencing using standard Illumina protocols
