Treatment protocol |
Each mouse was anesthetized, transcardially perfused with saline, and hippocamppus was isolated quickly, preserved at –80°C until RNA preparation.

Each mouse was maintained in SPF condition and sacrificed at 18 months of age.

GeneChip® WT PLUS Reagent Kit (Affymetrix) was used to generate amplified and biotinylated sense-strand DNA targets from expressed transcripts (100 ng of total RNA).

Hybridization protocol |
Following fragmentation, 5.5 µg of cDNA were hybridized for 17 hr at 45℃ on Affymetrix Mouse Gene 2.0 ST Array. GeneChips were washed and stained in the Affymetrix GeneChip Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the Affymetrix GeneChip® Scanner 3000 7G.
