Treatment protocol |
All experimental hens (35-40 weeks of age) were obtained from Wuhan 458 Xintianmu Agricultural Science and Technology Development Co. Ltd. (Hubei, China). For the androgen receptor-related drug experiment, chickens were randomly divided into 7 groups (10 hens/group). The androgen receptor antagonist-treated groups (3 groups) were fed 5 mg, 10 mg, and 15 mg of bicalutamide (AbMole, M1960) respectively, and the androgen agonist-treated groups (3 groups) were fed 1 mg, 10 mg, and 100 mg of mestanolone (AbMole, M3748) respectively, and the blank group (1 group) was fed with water as control. All drugs were fed for 4 days, and follicles were collected after 1 day of drug withdrawal. All hens were freely fed and watered.

Granulosa cells and theca cells were isolated from fresh follicular tissue of hens and were sent to Novogene Co., Ltd. (Beijing, China) for RNA extraction, library construction, and sequencing. The sequencing library was generated under the instruction manual with NEB Next UltraTM RNA Library Prep Kit for Illumina (NEB, USA). Obtained cDNA libraries were sequenced on an Illumina Nova platform and generated paired-end reads.
