Animals were euthanised with pentobarbital sodium (euthatal) and intracardially perfused with ice-cold PBS. Carotid bodies were excised from the common carotid artery bifurcation under the dissecting microscope and immediately snap-frozen in liquid nitrogen. Tissue was homogenized using QIAzol lysis reagent. Total RNA was extracted using phenol:chloroform phase separation method and isolated using ZYMO direct-zol RNA microprep kit following manufacturers instructions. Illumina TruSeq® Stranded mRNA library preparation kit (20020594, Illumina)

Original .fastq files were inspected using FastQC. As all samples passed initial QC no further trimming was applied .fastq were aligned to the rat reference genome (Rn6) using RNA-STAR mapper Reads were counted using featureCounts at a gene level Differential expression analysis was performed using DEseq2 Genome_build: Rn6 Supplementary_files_format_and_content: Matrix table with raw gene counts for every gene and every sample
