Treatment protocol The primary granulosa cells transfected with AR-specific siRNA (AR-siRNA) or NC nonsense siRNA (NC-siRNA) using Lipofectamine 3000 (Invitrogen Life Technologies, Carlsbad, CA, USA) according to the manufacturer’s instructions, respectively.
Growth protocol The primary granulosa cells were pre-cultured with Medium 199 (Gibco, USA) and 5% FBS (Gibco, USA) overnight (16h).
Extracted molecule polyA RNA
Extraction protocol Total RNA was extracted from primary granulosa cells using TRIzol reagent (Invitrogen, USA) following the manufacturer’s protocol.
The sequencing library was generated under the instruction manual with NEB Next UltraTM RNA Library Prep Kit for Illumina (NEB, USA). Obtained cDNA libraries were sequenced on an Illumina Nova platform and generated paired-end reads.

Data processing Reads were filtered by removing adapters, low quality reads and unknown bases.
Clean reads were mapped to Gallus_gallus-5.0 of Ensembl using HISAT2 v2.1.0.
Genes expression levels were quantified by a software package: stringtie v1.3.4d.
Differentially expressed genes were calculated by Deseq2 v1.26.0.
Genome_build: Gallus_gallus-5.0
Supplementary_files_format_and_content: Tab-delimited gtf files include "cov", "FPKM", "TPM" values for each Sample, which generated by stringtie.
