Treatment protocol |
Sample 1,2,3,7,8,9,13,14,15,19,20,21 were grown in normal growth conditions without any stress in green house. While sample 4,5,6, and 10,11,12 were subjected to water stress by with holding water for 7 days. Sample 16,17,18 and 22,23,24 were subjected to 40C for 60 min in growth chamber befor sample collection.

Trizol extraction of total RNA was performed according to the manufacturer's instructions.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Tomato Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

Scan protocol |
GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G3000.
