Extraction protocol |
Total RNA was isolated from MEF and Lymphoma samples using RNA Trizol per the manufacturer’s protocol and Qiagen RNAeasy purification. RNA was hybridized to Affymetrix Mouse 430A 2.0 arrays, and CEL files were preprocessed using Robust Multichip Average (RMA).

RNA of interest is converted into cDNA, then amplified and labeled with biotinylated nucleotides by in vitro transcription to produce biotinylated cRNA.

Hybridization protocol |
The cRNA is hybridized to a microarray chip. The bound cRNA is stained with fluorescent streptavidin.

Scan protocol |
The microarray chip is then scanned using a laser, and the positions and intensities of the fluorescent emissions are captured. These measurements provide the basis of subsequent biostatistical analysis.
