Extraction protocol Flash Frozen forebrains were extracted in nuclei extraction buffer, labelled with NeuN for 1hr then sorted with FACS. NeuN+ nuclei were pelleted, then split into ~200K aliquots for ChIP-seq. Nuclei were sonicated into 300bp fragments, then H3K4me3 or H3K27ac ChIP was performed.
Nugen Ovation Ultralow V2 library preparation - amplification for 17 cycles (H3K27ac), or 18 cycles (H3K4me3) 0347 V2 1-96/0344 V2 1-16.
