Genomic DNA (gDNA) was extracted from 1-month-old Arabidopsis thaliana rosette leaves with the DNeasy plant mini kit (Qiagen, cat. no. 69104) per the manufacturer’s instructions. Libraries were produced with the Ultra II DNA Library prep kit according to manufacturer’s instructions (New England Biolabs, cat. no. E7645L). Bisulfite conversion of DNA was carried out according to manufacturer’s protocol (Zymo, EZ DNA Methylation Lightning Kit, cat. no. D5046). DNA was converted twice to ensure complete bisulfite conversion of unmethylated cytosine.
