The posterior part of the embryo (immediately below the forelimb pit) was cut off and transferred separately to a 600 uL plastic tube containing 15 uL of RNAlater (Ambion).

Embryos were removed from pregnant dams at embryonic day 9.5. At E 9.5, the average embryo was āCā shaped and had 21.3 pairs of somites and neurul tube closure was complete with the exception of the posterior neuropore.
