Treatment protocol |
After 3 day culture in vitro, cells were harvested and lysised with 300 ul lysis buffer from the MirVana RNA Isolation kit.

Growth protocol |
Control and STAT-deficient naïve CD4+CD44-CD62L+ T cells were isolated and sorted on the flow cytometer. Cells from STAT3 WT and STAT3 KO were cultured for 72h with platebound anti-CD3, anti-CD28 (10 mg/ml each), IL-6 (20 ng/ml) TGF-beta (2 ng/ml) , anti-IL-2 (10 ug/ml) and anti-IL-4 (10 mg/ml) for 3 days. Cells from STAT5 WT and STAT5 KO were cultured for 72h with platebound anti-CD3, anti-CD28 (10 mg/ml each), IL-6 (20 ng/ml) TGF-beta (2 ng/ml) , IL-2 (100IU/ml) and anti-IL-4 (10 mg/ml) for 3 days.
