Extraction protocol CAR T cells are manufactured and injected into tumor-bearing mice. The tumors are dissected on day 8 post injection and analyzed. The input cells are also analyzed.
We followed the 10x Genomics instructions for library generation.

Data processing Raw scRNA-seq fastq files from sequencing were processed using CellRanger (v.6.1.2, 10X Genomics) software suite with corresponding human GRCh38 reference. Cell barcodes were determined based on the distribution of UMI counts, and filtered gene-barcode matrix was generated by CellRanger (v.6.1.2).
