Growth protocol Balb/c mice were infected with wild-type parasitesi or mutant parasites with binding sites of AP2-Sp muteted in the upstream of the AP2-Sp gene (AP2-Sp_cicmut). Infected mice were subjected to mosquito bites. Fully engorged mosquitoes were maintainedat 20 ℃. Midguts were disected 14 days after infective blood meal (oocysts/oosyst sporozoites). Salivary glands were disected 24 days after infective blood meal (salivary gland sporozoites).
Extracted molecule total RNA
Extraction protocol Dissected midguts were soaked in RNA Later solution until finishing the dissection. RNA extraction was performed using Isogen 2 (Nippon gene) according to the manufacturer’s protocol. Dissected midguts were lysed in lysis solution in the kit using Tissuelyser system (Quiagen). Total RNA was extracted using the ISOGEN II (NIPPON GENE, Japan) according to the manufacturer’s protocols.
Libraries were constructed from total RNA using KAPA mRNA Hyperprep kit (KAPA Biosystems), and sequencing was performed with Illumina NEXTseq sequence (Single read - illumina NextSeq 500),

Description transcriptome of salivary gland sporozoites
Data processing Reads were mapped on the P. berghei genome by HISAT2. Parameters of HISAT2 were set to default except for the option “--max-intron” set to 1000. Mapping data were analyzed by Feature counts software.
