Extraction protocol |
High molecular weight RNA (Ambion - miRvana) was polyA+ selected (Ambion - PolyApurist). We then fragmented the polyA with RNA fragmentation reagents (Ambion) and treated the RNA with T4 polynucleotide kinase. These treated RNAs were then ligated to a 3' adapter (IDT-Linker-1 or IDT-Linker-2). These 3'-captured fragmented RNAs were size selected and ligated to a barcoded 5' adapter, reverse transcribed, amplified and sequenced.
