Treatment protocol |
Mice were subcutaneous injected with a single dose of isoproterenol (ISO, 5 mg/kg body weight) or vehicle saline (10 ml/kg body weight). Hearts and vessels were harvested and immediately frozen in liquid nitrogen.

Extraction protocol |
Weigh out tissue and cut into cubes. Prepare protein extraction reagent containing inhibitors. Add ice-cold Protein Extraction Reagent containing protease inhibitors. Grind in a blender on low power using 30-second bursts until fully homogenized. Allow material to cool for 1 minute between bursts. Centrifuge the lysate at 14,000xg for 15 minutes and transfer the supernatant to a new tube for further analysis.

Hybridization protocol |
Cytokine arrays were used according to the manufacturer’s instructions. All sample measurements were performed in duplicate. The array plate were washed, incubated with a biotin-conjugated beads for half an hour, washed again, and developed for half an hour with PE-conjugated streptavidin.

Scan protocol |
Driven by the flowing sheath fluid, the microspheres pass through the red and green laser successively in a single row. The red laser is used to determine the fluorescence code of the microspheres, and the green laser is used to measure the fluorescence intensity of the molecules reported by the Luminex X-200.

Data processing |
By comparing the signal intensities, relative expression levels of cytokines can be made. The intensities of signals can be quantified by densitometry.
