Extraction protocol RNA was extracted using RNAmagic after manufacturer's protocol (Bio-Budget Technologies GmbH, Germany). cDNA synthesis was performed after Hagemann-Jensen et al. 2020 with minor modifications. Due to low cDNA amount whole length cDNA was amplified via PCR by controlling cycle number to avoid overamplification with LA Taq polymerase (TAKARA BIO INC., Japan).
Library preparation was done adopted to Picelli et al. 2014. The samples were equimolar pooled and quantified (with Agilent Bioanalyzer, DNA 7500). Sequencing was done on the NextSeq 500 (Illumina) using the High Output Kit v2.5 (75 cycles)

Differential expression for RNA-Seq was perfomed compairing to a control group (WholeAdults, Larval Trachea )
Assembly: assembly Release 6 plus ISO1 MT
