Treatment protocol |
Rat were exposed normoxia or hypoxia (10% oxygen) for 21d, freshly lung tissues from rats were isolated and perfused with PBS.

Extraction protocol |
Trizol extraction of total RNA was performed according to the manufacturer's instructions.The purity and concentration of total RNA samples were determined with NanoDrop ND-1000.

Total RNA from each sample was treated with Rnase R to enrich circular RNA. The enriched circular RNA was then amplified and transcribed into fluorescent cRNA utilizing ramdom primer according to Arraystar Super RNA Labeling protocol (Arraystar, Inc.).

Hybridization protocol |
The labeled cRNAs were hybridized onto the Arraystar Human circRNA Arrays V2 (8x15K, Arraystar), and incubated for 17 hours at 65°C in an Agilent Hybridization Oven.

After washing, slides were scanned with the Agilent Scanner G2505C.

Gene expression of rat lung tissue after hypoxia-treated for 21d
|
Data processing |
Agilent Feature Extraction software (version 11.0.1.1) was used to analyze acquired array images. Quantile normalization and subsequent data processing were performed using the R software limma package. Differentially expressed circRNAs with statistical significance between two groups were identified through Volcano Plot filtering. Differentially expressed circRNAs between two samples were identified through Fold Change filtering. Hierarchical Clustering was performed to show the distinguishable circRNAs expression pattern among samples.
