Single cell cDNA libraries were prepared using oligo(dT) primed cDNA synthesis followed by PCR amplification of cDNAs, as described in previously published methods (Hanchate et al, 2015, Science, 1251-5; Malnic et al, 1999, Cell, 96:713.) Libraries were prepared for sequencing using the TruSeq DNA sample prep kit following the manufacturer's instructions (Illumina)

Data processing |
Image analysis and basecalling was performed using Illumina's Real Time Analysis v1.8.66.3 software, followed by demultiplexing of indexed reads and generation of FASTQ files with Illumina's bcl2fastq-1.8.4 software. Sequenced reads were aligned to iGenome's UCSC mm10 using TopHat aligner v2.1.0. Cufflinks (cuffquant followed by cuffnorm) were used to generate gene abundances Assembly: mm10 Supplementary files format and content: tab-delimited file containing FPKM normalized gene counts per sample Supplementary files format and content: tab-delimited file containing FPKM normalized GPCR counts per sample
