Treatment protocol |
MPFR and TPFR were established through long-term treatment with 100 nM fulvestrant and 200nM palbociclib over a period of 2-4 months.

Growth protocol |
MPFR cells were cultured in phenol red-free Dulbecco’s modified Eagle medium (D-MEM)/F-12 (Gibco) supplemented with 1% heat-inactivated fetal bovine serum (FBS, Gibco), 2 mM glutamine (Gibco) and 6 ng/ml insulin (Sigma-Aldrich) supplemented with 200 nM CDK4/6i and 100 nM fulvestrant. TPFR cells were cultured in Roswell Park Memorial Institute (RPMI) 1640 media without phenol red (Gibco, ThermoFisher Scientific, Waltham, MA, USA), supplemented with 5% FBS, 2 mmol/L glutamine, 8 µg /mlL insulin and 200 nM CDK4/6i and 100 nM fulvestrant.

Extraction protocol |
Total RNA was extracted and purified with RNeasy Mini kit (Qiagen) according to the manufacturer´s instructions.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on human transcriptome array 2.0 (HTA). GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

Scan protocol |
GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G2500A.
